The nucleocapsid protein (N-protein) is a structural protein that binds to the coronavirus RNA genome, thus creating a shell (or capsid) around the enclosed nucleic acid. The N-protein also interacts with the viral membrane protein during viral assembly, assists in RNA synthesis and folding, plays a role in virus budding, and affects host cell responses, including cell cycle and translation. Coronavirus N protein is required for coronavirus RNA synthesis and has RNA chaperone activity that may be involved in template switch. Nucleocapsid protein is the most abundant protein of coronavirus. During virion assembly, N protein binds to viral RNA and leads to the formation of the helical nucleocapsid. Nucleocapsid protein is a highly immunogenic phosphoprotein also implicated in viral genome replication and in modulating cell signaling pathways. Because of the conservation of the N protein sequence and its strong immunogenicity, the N protein of coronavirus is chosen as a diagnostic tool.

The binding activity of HA600005 with Recombinant SARS-CoV-2 Nucleocapsid protein.
Immobilized Recombinant SARS-CoV-2 nucleocapsid protein at 1 μg/ml overnight at 4℃. Then blocked with 1% BSA for 1 hour at 37℃, and incubated with the primary antibody (HA600005) for 1 hour at 25℃. The EC50 of HA600005 is 47.22 ng/ml. -
This antibody will detect SARS-CoV-2 Nucleocapsid protein in ELISA pair set (Clone: # A4G9-HRP). In a sandwich ELISA, it can be used as Detect antibody when paired with (Cat: # HA600007).

Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark. Detector: HA600005, SARS-CoV-2 Nucleocapsid protein Rabbit mAb [A4G9]
Elisa assay was performed by coating wells of a 96-well plate with 100 µl per well of capture antibody (HA600008) diluted in carbonate/bicarbonate buffer, at a concentration of 2 µg/mL overnight at 4℃. Wells of the plate were washed, blocked with 150 µl 0.05% tween-20 1% BSA blocking buffer, and incubated with serial diluted Recombinant SARS-CoV-2 Nucleocapsid protein protein (HAg2002) starting from 4,000,000 pg/ml to 0 pg/ml and detect antibody (HA600005, Biotin, 0.5 µg/ml) for 1 hour at 30℃ with shaking. Then the plate was washed and incubated with 100 µl per well of SA-HRP for 0.5 hour at 30℃ with shaking. Detection was performed using an Ultra TMB Substrate for 10 minutes at room temperature in the dark.
